ABSTRACT

Salmo trutta fario is the most important fish species of family Salmonidae inhibited in cool waters all over the world including the nortern parts of Pakistan. This study was aimed to find out the prey selection and feeding habits of the species. One hundred and eighty five Salmo trutta fario were captured from March 2016 to April 2017. Feeding intensity as represented by a fullness index (FI), changing with various length groups and season. Out of 185 Brown trout 24 fish stomaches were found empty. The most important and common food items of brown trout were Brachycentridae, Blepharocera, Hydropsychidae, ephemerella spp. Kruskal Wallis H-test were applied on feeding intensity groups consisting of three month group. The test statistic for K.W-H-test were (H=8.13 with df =3) had a p-value of 0.043 < (alpha=0.05 indicates favor of the alternative hypothesis of at least one difference among the feeding intensity groups. The linear relationship of N with index of relative importance and %IRI denoted by least square regression line (N = 35.2 + 22.1%IRI), shows that for 22 prey there is 1% change in IRI. The relationship between total length (mm) and fish body weight (gm) is expressed by Pearson correlation coefficient (r = 0.976), showed that total length (mm) and fish body weight (gm.) is highly correlated. Descriptive statistics are used for the stomach fullness, which shows that feeding intensity was recorded higher from March to May. A total of 2289 preys was recorded including the most common were; Trichoptera, Hydropsychidae, Brachycentridae, diptera, blepharocera, ephemerala, chironomida, honey bees, grass hopper, locust, trout egg, trochanter, plant tissue, stones were retrieved from the gut contents of brown trout. According to index of relative importance IRI% four preys represents major components of the diet. The highest IRI% was recorded in Brachycentridae (39.38%), followed by blepharocera (13.23%), Hydropsychidae (10.76%) and ephemerella spp (8.28%). The relationship between IRI and FO is (r =0.556) is moderately correlated with coefficient of determination (r2=0.31). This study will help in the development of an artificial diet for the species for better growth performance in captivity.

ABSTRACT

ABSTRACT The aquaculture industry aims to find new strategies to replace fishmeal in formulated diet-sources with high attraction and palatability and high biological value to replace conventional sources. Due to the nutritional potential of animal by-products, technologies have been adopted for their use, such as bioconversion techniques by enzymatic hydrolysis. Consequently, the objective of this study was obtaining and evaluating a hydrolyzed protein concentrate of animal waste (CPHRA, by its Spanish acronym) in diets for Piaractus brachypomus. Viscera from poultry by-products was evaluated by using formic acid and a 20% trout viscera to promote the catalyst at lower pH (3,66). The resulting product was used in feed formulation to determine the Apparent Digestibility Coefficient (ADC) of energy and nutrients, using a control diet (T0) and three diets with 10%, 20%, and 30% CPHRA (T1, T2 y T3). One hundred and twenty juveniles of hybrids of cachama blanca with an average weight of 145 ± 3,0 g were fed in triplicate groups. The CPHRA presented high protein content and significant differences (p < 0,05) were evidenced for the ADCs of dry matter, crude protein, crude fat, crude energy, and ash absorption, calcium and phosphorus. The ADCs for nutrients and energy were above 80%. Accordingly, the hydrolyzed protein concentrate from poultry waste has great potential as a protein and energy alternative of high nutritional value in fish feeding.

ABSTRACT

Abstract Studies in Salvelinus alpinus, Arctic charr, indicate that it has a low capacity to hyposmorregulatory or adaption to sea in winter periods in Arctic waters. The investigation finds to determinate the rank optimum of salinity to can cultivate this species at Chile. The weight adequate was determined to join on the sea by analysis of gill Na+, K+-ATPase activity, that it was found between the ranks 80-130 g, with 14.5 U/mg. It underwent evaluation of fish growth of 72 g salinities from 0 (control), 18, 25 and 33 g/L (sea water) for 94 days. The results indicate that the largest increases were obtained in brackish water. T18 g/L and T25 g/L achieved growth of 25% and 19% on day 94 and term sampling respectively. It is important to mention that the 8% that survived in seawater introduced percentages growth 16.6% equivalent to brackish water and control. These results suggest that Salvelinus alpinus can grow in seawater, with levels of Na+, K+-ATPase similar to those submitted by Salmo salar with a weight not less than 80 g.

ABSTRACT

The aim of this study was to determine the food webs structure of a large Patagonian river in two river sections (Upstream and Midstream) and to evaluate isotopic overlap between native and introduced species. We used stable isotope analyses of δ15N and δ13C and stomach content. The Upstream section had a more complex food webs structure with a greater richness of macroinvertebrates and fish species than Midstream. Upstream basal resources were dominated by filamentous algae. Lake Trout were found to have a higher trophic position than all other fish species in that area although, the most abundant fish species, were Rainbow Trout. Depending on the life stage, Rainbow Trout shifted from prey to competitor/predator. In the Midstream section, the base of the food webs was dominated by coarse particulate organic matter, and adult Rainbow Trout had the highest trophic level. Isotopic values changed among macroinvertebrates and fish for both areas. The two most abundant native and invasive species — Puyen and Rainbow Trout — showed an isotopic separation in Midstream but did not in Upstream areas. The presence of invasive fish that occupy top trophic levels can have a significant impact on native fish populations that have great ecological importance in the region.(AU)

ABSTRACT

BACKGROUND: Polydeoxyribonucleotide (PDRN) influencing cellular growth and differentiation is recognized to promote wound healing by stimulating tissue repair. Although PDRN can be extracted from human placentas, PDRN medications have recently been extracted from the semen of trout (Oncorhynchus mykiss) and salmon (Oncorhynchus keta). The present study was designed to evaluate the wound healing effects of O. keta-derived PDRN for injection (Rejuvenex) and PDRN cream (Rejuvenex Cream) in comparison with those of O. mykiss-derived PDRN injection (Placentex). METHODS: Full-thickness skin defects were made on the back of mice (n=60). The mice were divided into the following four groups according to the dressing used for the wounds: O. mykiss-derived PDRN injection group, O. keta-derived PDRN injection group, O. keta-derived PDRN cream group, and normal saline soaked dressing group (control group). We analyzed the gross findings, wound sizes, histological findings, immunohistochemistry and enzyme-linked immunosorbent assays for the groups immediately after the treatment, and again after 4, 7, and 10 days of treatment. RESULTS: The wound healing effects were the greatest in the O. keta-derived PDRN injection and O. mykiss-derived PDRN injection groups, which showed similar scores, followed by the O. keta-derived cream and normal saline soaked dressing groups. CONCLUSION: The injection of PDRN extracted from O. keta was found to be as effective at healing full-thickness skin defects as the O. mykiss-derived PDRN injection, which is currently used in the clinic. Moreover, the O. keta-derived PDRN injection was also found to reduce the time required for wound healing.

ABSTRACT

The infectious pancreatic necrosis (IPNV) is the causative agent of an acute illness well characterized in salmonids worldwide. Clinical signs and mortality rates are dependent on several factors such as the viral dose, the age of the fish, the water temperature, among others. An experimental study was conducted to measure the effect of temperature on the gene expression profile of IFN-1(α), STAT-1 and Mx-1 in rainbow trout fry, exposed to IPNV. Fry (n=198) were exposed at 8, 12 and 16°C, and samples were taken for 21 days to determine the virus titer and gene expression. In the first 11 days the greatest viral titer was recorded at 8°C compared with the values obtained at 12 and 16°C. At 8°C, there was a significant increase on day 4 of mRNA Mx-1 (t-test, p<0.05), time in which the viral titer began to decrease. Furthermore, as the viral titer increased, STAT-1 and Mx-1 (r=0.91) and (r=0.96) increased, respectively. The animals were able to recover from day 4 from some of the symptoms of IPN. Clinical disease was developed only in fish exposed to 12°C and all died between days 6 and 14, despite the highly significant increase shown in the average expression level of Mx-1, compared with the values recorded at 8°C and 16°C (Tukey, p<0.0001). Additionally, the expression profiles of IFN-1(α) and STAT-1 decreased completely (~0.016) and (~0.020 times) on day 7. The highest expression level of IFN-1(α), occurred at 16°C (Tukey, p<0.0005). Fry exposed at 16°C were normal during the experiment. IFN-1(α) possibly generated a protector effect from day 2 when they showed a significant expression increase compared with the results at 8°C and 12°C (t-student, p<0.0001); however, STAT-1 was not significantly affected by temperature, although the highest average expression value was recorded at 16°C. Our research supports the expression of relevant anti-viral response genes as IFN-1(α), STAT-1 and Mx-1 are physiologically modulated by the water temperature, directly influencing the development of the IPN disease in rainbow trout.

ABSTRACT

Outbreak and development of yersiniosis in rainbow trout farms in Iran has caused a serious problem over the last years. The purpose of this study was to evaluate the efficacy of immersion vaccination with Yersinia ruckeri in rainbow trout. Prior to antigen preparation, the phenotypic, molecular and serological features of a number of Yersinia ruckeri isolates obtained from affected trout farms were studied. The virulent of these isolates were then evaluated using intra peritoneal injection route. Trout were vaccinated by immersion route [3 min at 12 °C] using Yersinia ruckeri bacterin of the virulent strains. The efficacy of vaccine antibody titer within 2, 4, 6, 8 and 10 weeks post vaccination were evaluated using relative percent survival. The phenotyping, serological and molecular studies have led to identification of 8 isolates of Yersinia ruckeri and all the isolates produced bands 409 bp, which is indicative of Yersinia ruckeri. In pathogenicity test 3 isolates caused above 50% mortality, while 5 isolates reached 16%. The RPS of vaccinated fish reached 72.7, 80, 80, 82.2 and 83.3% within 2, 4, 6, 8 and 10 weeks post vaccination, respectively. In the other words, the mortality level in vaccinated groups was in range of 10-20% within 10 weeks post vaccination, while those of control group was in range 56.7 - 73.3% [p<0.05] .The lowest and the highest antibody titers in immunized groups were 32 +/- 4.50 and 164.57 +/- 9.37 respectively, obtained after 4 and 10 weeks of immunization, whereas the control group had no measurable titer of antibody. The results of this study clearly show that this vaccine can remarkably protect the trout from yersiniosis outbreaks inside Iran

ABSTRACT

Today, uses of natural substances are preferred to chemical and synthetic compounds for growth and immune enhancement of aquatic organisms. The aim of this research was to evaluate the effect of Saccharomyces cerevisiae extract and hydrolyzed powder on growth parameters and resistance of rainbow trout against temperature and hypoxia stresses with a bacterial challenge. For this purpose six hundred rainbow trout [50 +/- 5 g mean weight] were purchased from a local fish farm and transferred to the laboratory. After acclimatization to the laboratory conditions the fish were randomly divided into four groups each with triplicate. The first group was fed with a commercial diet [control] while the other groups received yeast extract, hydrolyzed powder and a mixture of them with the commercial diet. Fish were cultured in a 300 l polyethylene tank for 60 days and growth parameters were analyzed at days 0 and 60. Also, at the end of study the fish were challenged with hypoxia and temperature stresses with an experimental bacterial infection. Results showed that the extract and hydrolyzed powder of Saccharomyces cerevisiae in combination significantly [p=0.021] improved rainbow trout growth parameters compared with the control. The results also proved that fish survival significantly increased following feeding with a mixture of the extract and hydrolyzed powder after thermal [69.55 +/- 4.59], hypoxia [96.55 +/- 5.34] and yersiniosis [49.11 +/- 2.18] challenge than the control. Dietary administration of yeast extract with mixture of hydrolyzed powder of Saccharomyces cerevisiae could improve rainbow trout growth parameters and increase the resistance against stresses and disease

ABSTRACT

BACKGROUND: Partial or complete necrosis of a skin flap is a common problem. Polydeoxyribonucleotide (PDRN) can be extracted from trout sperm and used as a tissue repair agent. The aim of this study was to investigate whether PDRN could improve the survival of random pattern skin flaps in rats. METHODS: Twenty-two male Sprague-Dawley rats were randomly divided into two groups: the PDRN treatment group (n=11) and the control group (n=11). Caudally pedicled random pattern skin flaps were elevated on their dorsal skin and resutured. The treatment group received daily intraperitoneal administration of PDRN (8 mg/kg/day), and the control group received fluid vehicle (NaCl 0.9%, 8 mg/kg/day) from day 0 to day 6. On day 7, the flap survival was evaluated and the harvested tissue surrounding the demarcation line of the necrotic area was stained with H&E, anti-rat vascular endothelial cell growth factor (VEGF) antibody, and PECAM-1/CD31 antibody. RESULTS: The average necrotic area of the flap in the PDRN group was significantly smaller when compared with that of the control group. Histologic and immunohistochemical evaluation showed that granulation thickness score and VEGF-positive staining cells were marked higher in the PDRN group than in the control group. PECAM-1/CD31-positive microvascular densities were significantly higher in the PDRN group when compared with the control group. CONCLUSIONS: This study confirms that PDRN improves the survival of random pattern skin flaps in rats. These results may represent a new therapeutic approach to enhancing flap viability and achieving faster wound repair.

ABSTRACT

There is a limitation to estimate vitamin B12 intake due to a lack of data on vitamin B12 content in many Korean foods. In this study, vitamin B12 content was determined in some seaweeds, fish, and shellfish and their product that are consumed in Korea using a modified microbioassay with Lactobacillus delbruecki ATCC 7830. Dried laver and dried seasoned and toasted laver contained very high levels of vitamin B12 (66.8 and 55.2-71.3 microg/100 g, respectively. Sea lettuce and seaweed fulvescene also contained high vitamin B12 content of 5.47-9.41 and 6.46-7.20 microg/100 g, respectively, whereas sea mustard and sea tangle contained low levels of vitamin B12; vitamin B12 was not detected in seaweed fusifome. Pacific saury, trout, sea-bass, or squid contained 12.01, 2.00, 0.49 and 2.33 microg vitamin B12/100 g, respectively. Ochellatus octopus, and naked sand lance contained 0.72-1.43 and 3.68 microg vitamin B12/100 g, respectively. Dried Alaska pollack contained 0.19-2.64 microg vitamin B12/100 g. Shellfish such as little neck clam and small ark shellfish contained high levels of vitamin B12 of 30.5-40.5 microg/100 g, and mussel and abalone contained 17.71 and 7.82 microg/100 g, respectively. Of unique Korean traditional fermented seafood products, salt-fermented products of squid (2.91 microg/100 g), clams (34.31 microg/100 g), Alaska pollack roe (9.98-12.02 microg/100 g), hairtail guts (4.58 microg/100 g) or small shrimp (0.58-1.55 microg/100 g), and fish sauce from anchovies (1.52-1.78 microg/100 mL), sand eel (0.22-0.24 microg/100 mL) or small shrimp (0.19-0.78 microg/100 mL) were analyzed. A few commercial brands of flying fish roe (0.73-1.73 microg/100 g), canned tuna (0.40 microg/100 g), and fried fish paste (0.25-0.69 microg /100 g) were also analyzed. In conclusion, vitamin B12 content in these foods, chosen considering the Korean food culture, should contribute to improve the present vitamin B12 food database. It may be helpful to estimate vitamin B12 intake more correctly than before, and provide additional information for dietary education related to vitamin B12 and meal management.

ABSTRACT

The use of probiotic bacteria has been suggested as an important strategy to accomplish reproductible outputs through biocontrol in cultivation systems for marine fish larvae and crustaceans. The bacterial flora in the larval gut originates from bacteria associated with the eggs, the water in the rearing tanks, and the live food. This study was aimed to determine the effect of commercial and Autochthonous probiotics on growth parameters and survival rate of Rainbow trout [Oncorhynchus mykiss] larvae. Trout larvae [85 +/- 4 mg] were fed diets of Biomar. The experimental diets were supplemented with three blends of probiotic bacteria including lactobacillus, commercal Bacillus and isolated sturgeon gut bacillus in three levels [4.30, 5.30 and 6.30 LogCFU/g of feed] and were fed by Rainbow trout [larvae in 9 experimental treatments. The control treatment was fed on nonsupplemented diet. The experiment was carried out in completely random design. At the end of the period the fishes were biometeried and tested by thermal, salinity, alkalinity and acidity stress. The highest specific growth rate, thermal growth coefficient and feed conversion efficiency were found in treatment Al. The treatments A1, A2, B1, B2, L1 andL3 had significant difference with control [p0.05]. The beter feed conversion ratio was obtained in treatment A1 and L3. Inchalleng test of thermal stress, maximum of survival time. was obtained in treatment B 1. The results of the challenge tests with stress of alkalinity and acidity indicated that the best survival time was in treatment Al and total experimeal treatments had significant difference with control [p<0.05]. No significant difference in salinity challenge test was observed between the treatments [p>0.05]

ABSTRACT

Determination of the accurate time of egg stripping after ovulation has been regarded as one of the most important factors in the successful reproduction of Caspian brown trout[Salmo trutta caspius]. To determine the best time for egg stripping after ovulation and over-ripening of the oocyte in the Caspian brown trout, the eggs of fish were retained in the coelomic cavity of females at 7 +/- 6°C over 40 days of post-ovulation[DPO]. Eggs were stripped at 10 day intervals in 4 stages and then were fertilized with the mixed milt of males. The obtained results showed that the eyeing rate decreased from 90.65 +/- 6.28% in newly ovulated eggs [0-10 DPO]to 0.67 +/- 1.34% in over-ripened eggs[30-4ODPO]. Also, the hatching rate decreased from 86.33 +/- 6.28% in newly ovulated eggs to 0.49 +/- 0.98% in over-ripened eggs. However, larval abnormalities remained constant for 30 days after ovulation. The histological study in the newly ovulated egg showed that the yolk consisted of homogenous tissue having the perivitelline space diameter with no considerable difference. At over-ripening stage, the yolk became hetero generous, while chorion diameter did not change. Also the perivitelline space diameter varied among different areas. The micropylar apparatus also showed no change during the over-ripening stage. The present study demonstrated that the best time to strip Caspian brown trout eggs post-ovulation was up to 10 DPO at 7 +/- 0.6°C. Same histological changes occurred in oocyte at over-ripening hut it showed no effect on the micropyle

ABSTRACT

Iran is one of the most important Rainbow trout [Oncorhynchus mykiss] producing countries in the world and fingering production is the key factor for this industry. The present experiment was conducted to determine the role of the coelomic fluid in artificial fertilization and to study the interactive effect of different brood fish coelomic fluid on fertilization process during 60 min storage at 4°C.The egg quality was evaluated through the rate of eyed and hatched eggs where these indices applied with and without coelomic fluid. In the first two trials, the rat of egg fertilization with and without coelomic fluid did not exhibit significant difference [p>0.05]. Eyeing rates were 97.1% and 97.6% for trial 1 and 99.1% and 97.7% for trial II. respectively. In next trial the eyeing and hatching rate were 88.7% and 86.8% for the eggs fertilized immediately alter stripping wheas for those fertilized 60 min. after integrated storage these values were 86.3% and 84.6% respectively [p>0.05]. we concluded that the coelomic fluid has no interative effect on artificial fertilization and the quality of egg fecundity is not influenced by the fertilization process during 60 min storage at 4°C

ABSTRACT

The aim of this study was to analyze the morphological variation of brown trout (Salmo trutta) in the Duero basin, an Atlantic river basin in the Iberian Peninsula, where a spatial segregation of two divergent lineages was previously reported, based on isozyme, microsatellite and mtDNA data. In these studies, two divergent pure regions (Pisuerga and Lower-course) and several hybrid populations between them were identified. Morphological variation was evaluated in 11 populations representative of the genetic differentiation previously observed in the Duero basin, using multivariate analysis on 12 morphometric and 4 meristic traits. A large differentiation between populations was observed (interpopulation component of variance: 41.8 percent), similar to that previously detected with allozymes and microsatellites. Morphometric differentiation was also reflected by the high classification success of pure and hybrid individuals to their respective populations, using multivariate discriminant functions (94.1 percent and 79.0 percent, respectively). All multivariate and clustering analyses performed demonstrated a strong differentiation between the pure regions. The hybrid populations, though showing large differentiation among them, evidenced an intermediate position between the pure samples. Head and body shape traits were the most discriminant among the morphometric characters, while pectoral rays and gillrakers were the most discriminant among the meristic traits. These results confirmed the high divergence of the brown trout from the Duero basin and suggest some traits on which selection could be acting to explain the spatial segregation observed.

ABSTRACT

PURPOSE: This study involved a population-based survey to provide evidence of public awareness of risk factors of gastric cancer and to investigate attitudes for the screening of gastric cancer in the South Korean population. MATERIALS AND METHODS: Using a nationwide random selection method, 2014 subjects were enrolled in the study between 5 September 2006 and 25 September 2006. RESULTS: In terms of the awareness of risk factors, awareness was scored as the percentage of the probability of developing gastric cancer when a subject had a particular risk factor. For the risk factors, stress ranked highest with a score of 73.5%, followed by chronic gastritis (score of 72.1%), gastric ulcer (score of 71.2%) and a previous gastrectomy history (score of 68.7%). Other factors included a diet of charred foods (score of 67.3%), alcohol use (score of 65.3%), salty diet (score of 65.1%), history of smoking (score of 64.3%) and Helicobacter pylori infection (score of 57.5%). Subjects believed that 60.4% of all gastric cancers were preventable by lifestyle modification and the subjects believed that regular screening could prevent 72.1% of all gastric cancers. However, 54% of subjects did not receive regular screening and the most common reason for not undergoing screening was a lack of symptoms. CONCLUSION: Public education about the risk factors of gastric cancer and of lifestyle modifications and the importance of regular screening regardless of the presence of symptoms should be emphasized to reduce gastric cancer mortality in South Korea.
